Pilot Comparison of 68Ga-RM2 PET and 68Ga-PSMA PET in Patients with Biochemically Recurrent Prostate Cancer
Objectives: Glu-NH-CO-NH-Lys-(Ahx)-[68Ga(HBED-CC)] (68Ga-PSMA) is a positron emission tomography (PET) tracer that can detect prostate cancer relapses and metastases by binding to the extracellular domain of PSMA. 68Ga-labeled DOTA-4-amino-1-carboxymethyl-piperidine-D-Phe- Gln-Trp-Ala-Val- Gly-His-Sta-Leu-NH2 (68Ga-RM2) is a synthetic bombesin receptor antagonist that targets gastrin-releasing peptide receptors (GRPr). We present pilot data on the biodistribution of these PET tracers in a small cohort of patients with biochemically recurrent prostate cancer (BCRPC). Methods: Seven men (mean age ± SD: 74.3±5.9 year-old) with BCRPC had both 68Ga-PSMA PET/CT and 68Ga-RM2 PET/MRI scans. The maximum standardized uptake value (SUVmax) and mean SUV (SUVmean) measurements were recorded in normal tissues and areas of uptake outside the expected physiologic biodistribution. Results: All patients had rising prostate-specific antigen (PSA) (mean±SD: 13.5±11.5) and non-contributory conventional imaging. 68Ga-PSMA had the highest physiologic uptake in the salivary glands and small bowel, with hepatobiliary and renal clearance noted, while 68Ga-RM2 had the highest physiologic uptake in the pancreas, with renal clearance noted. Uptake values uptake outside the expected physiologic biodistribution were not statistically different between 68Ga-PSMA and 68Ga-RM2; however, 68Ga-PSMA localized in a lymph node and seminal vesicle in a patient with no abnormal 68Ga-RM2 uptake. Abdominal periaortic lymph nodes were more easily visualized by 68Ga-RM2 in two patients due to lack of interference by radioactivity accumulation in the small intestine. Conclusion: 68Ga-PSMA and 68Ga-RM2 have distinct biodistribution in this small cohort of patients with BCRPC. The findings here indicate that additional work is needed to understand the expression of PSMA and GRPr in different types of prostate cancer.

A Population Based Gaussian Mixture Model Incorporating 18F-FDG-PET and DW-MRI Quantifies Tumor Tissue Classes
The aim of our study was to create a novel Gaussian Mixture Modeling (GMM) pipeline in order to model the complementary information derived from 2-deoxy-2-(18F)-fluoro-D-glucose (18F-FDG)-Positron Emission Tomography (PET) and Diffusion Weighted Magnetic Resonance Imaging (DW-MRI) measurements to separate the tumor microenvironment into relevant tissue compartments and follow the development of these compartments in a longitudinal manner. Methods: Serial 18F-FDG-PET and Apparent Diffusion Coefficient (ADC) maps derived from DW-MR images of NCI-H460 xenograft tumors were co-registered and a population based GMM was implemented on the complementary imaging data. The tumor microenvironment was segmented into three distinct regions and correlated with histological stainings. An analysis of covariance (ANCOVA) was applied to gauge how well the total tumor volume was a predictor for the ADC and 18F-FDG, or if ADC was a good predictor of 18F-FDG for average values in the whole tumor or average necrotic and viable tissues. Results: The co-registered PET/MR images were in excellent agreement with histology, both visually and quantitatively, and allowed for validation of the last time point measurements. Strong correlations were found for the necrotic (r = 0.88) and viable fractions (r = 0.87) between histology and the clustering. The GMM provided probabilities for each compartment with uncertainties expressed as a mixture of tissues in which the resolution of scans was inadequate to accurately separate tissues. The ANCOVA suggested that both ADC and 18F-FDG in the whole tumor (P = 0.0009, P = 0.02) as well as necrotic (P = 0.008, P = 0.02) and viable (P = 0.003, P = 0.01) tissues were a positive, linear function of total tumor volume. ADC proved to be a positive predictor of 18F-FDG in the whole tumor (P = 0.001) and necrotic (P = 0.02) and viable (P = 0.0001) tissues. Conclusion: The complementary information of 18F-FDG and ADC longitudinal measurements in xenograft tumors allows for segmentation into distinct tissues when utilizing the novel GMM pipeline. Leveraging the power of multi-parametric PET/MR imaging in this manner has the potential to take the assessment of disease outcome beyond the Response Evaluation Criteria in Solid Tumors and could provide an important impact to the field of precision medicine.

Multiparametric analysis of the relationship between tumor hypoxia and perfusion using 18F-FAZA and 15O-H2O PET
18F-fluoroazomycin-arabinoside (18F-FAZA) is a positron emission tomography (PET) tracer of tumor hypoxia. However, as hypoxia often is associated with decreased perfusion, delivery of 18F-FAZA might be compromised, potentially disturbing the association between tissue hypoxia and 18F-FAZA uptake. The aim of this study was to gain insight in the relationship between tumor perfusion and 18F-FAZA uptake. Methods: Ten patients diagnosed with advanced non-small cell lung cancer (NSCLC) underwent subsequent dynamic 15O-H2O and 18F-FAZA PET scans with arterial sampling. Parametric images of both 15O-H2O derived perfusion (TBF) and volume of distribution (VT) of 18F-FAZA were generated. Next, multiparametric classification was performed using lesional and global thresholds. Voxels were classified as low or high TBF and 18F-FAZA VT, respectively. Finally, by combining these initial classifications, voxels were allocated to four categories (lowTBF-lowVT, lowTBF-highVT, highTBF-lowVT and highTBF-highVT). Results: A total of thirteen malignant lesions were identified in the ten patients. Average TBF and 18F-FAZA VT across all lesions were 0.45±0.20 mL·cm-3·min-1 and 0.94±0.31 mL·cm-3, respectively. The average of all lesional median values were 0.37±0.15 mL·cm-3·min-1 and 0.85±0.18 mL·cm-3 for TBF and 18F-FAZA VT, respectively. Multiparametric analysis showed that classified voxels were clustered rather than randomly distributed. Several intralesional areas could be identified where 18F-FAZA VT was inversely related to TBF. On the other hand, there were also distinct areas where TBF as well as 18F-FAZA VT were decreased or increased. Conclusion: The present data indicate that spatial variation of 18F-FAZA uptake is not necessarily inversely related with TBF. This suggests that decreased TBF may result in flow limited delivery of 18F-FAZA. Areas with both high 18F-FAZA uptake and high TBF suggest that high 18F-FAZA uptake, possibly suggesting hypoxia, may occur in regions despite having high TBF. In conclusion, multiparametric evaluation of the spatial distribution of both TBF and 18F-FAZA uptake may be helpful for understanding the 18F-FAZA signal.

Behçet's syndrome (BS) is associated with chronic inflammation and endothelial dysfunction. Although there have been extensive investigations on neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), and mean platelet volume (MPV) in many diseases, their roles in BS is unclear. The purpose of the present study was to evaluate NLR, PLR, and MPV levels in BS patients and explore their clinical significance. The study included 254 patients with BS and 173 healthy individuals. Age, sex, age of onset, duration of disease, smoking, Behçet activity score, total white blood counts, neutrophil, platelet, and T lymphocyte counts of the patients were recorded. White blood cell (WBC), neutrophil, platelet, NLR, and PLR were significantly higher in patients with BS when compared with healthy controls (all p < 0.001). Lymphocyte counts and MPVs of the BS group were not statistically different from healthy controls (all p ˃ 0.05). In the BS group, PLR and MPV were significantly different among the three severity groups (p = 0.037 and p = 0.016, respectively). We showed that any laboratory markers were not associated with joint, eye, central nervous system, large vessel, or gastrointestinal involvement in BS. NLR was shown to be an independent factor for BS by multivariate analysis. We suggest that NLR can be considered to be a diagnostic criterion of BS given the support of the findings from larger prospective studies.

The Italian Diabetes and Exercise Study 2 (IDES-2): a long-term behavioral intervention for adoption and maintenance of a physically active lifestyle
Background: Physical activity (PA)/exercise have become an integral part of the management of type 2 diabetes mellitus (T2DM). However, current guidelines are difficult to put into action in this population due to a number of barriers, especially the lack of acceptable, feasible, and validated behavioral intervention strategies. The present manuscript reports the rationale, study design and methods, and design considerations of the Italian Diabetes and Exercise Study (IDES)-2, a randomized controlled trial testing the efficacy of a behavior change strategy in increasing total daily PA and reducing sedentary time (SED-time) in patients with T2DM.Methods/DesignStarting 7 January 2014, the IDES_2 began enrolling 300 patients with known T2DM of at least 1-year duration in three tertiary referral outpatient Diabetes Clinics in Rome. Additional requirements are age 40 to 80 years, body mass index 27 to 40 kg/m 2 , sedentary lifestyle, and physically inactive for at least 6 months, ability to walk 1.6 km without assistance, and eligibility after cardiovascular evaluation. Patients are randomized by center and within each center, by age and type of diabetes treatment to either the intervention or the control group. Patients in the intervention (INT) group (n = 150) receive theoretical and practical exercise counseling consisting of aggregated behavior change techniques (one individual theoretical counseling session plus eight twice-a-week individual theoretical and practical exercise counseling sessions) once a year for 3 years. Patients in the control (CON) group (n = 150), receive standard care, including general physician recommendations for daily PA. The primary outcomes are total daily PA and SED-time, as measured objectively by the use of an accelerometer. Secondary outcomes include physical fitness, modifiable cardiovascular risk factors, musculoskeletal disturbances, well-being/depression, and health-related quality of life.DiscussionThe behavioral intervention strategy tested in the IDES_2 is based on solid theoretical grounds and uses several behavioral change techniques, two factors which were found to improve effectiveness of behavioral intervention. In addition, physicians and exercise specialists have been specifically trained for counselling/prescribing and supervising PA/exercise, respectively, in subjects suffering from metabolic disorders. Finally, the large sample size, the long study duration, and the objective measurement of PA allow statistically significant and scientifically robust conclusions to be drawn on the feasibility and efficacy of this intervention in T2DM patients.Trial registrationClinicalTrials.gov; NCT01600937; 10 October 2012.

Renewable energy assessments for resort islands in the South China Sea were conducted that involves the collection and analysis of meteorological and topographic data. The meteorological data was used to assess the PV, wind and hydropower system potentials on the islands. Furthermore, the reconnaissance study for hydro-potentials were conducted through topographic maps in order to determine the potential sites suitable for development of run-of-river hydropower generation. The stream data was collected for 14 islands in the South China Sea with a total of 51 investigated sites. The data from this study are related to the research article "Optimal combination of solar, wind, micro-hydro and diesel systems based on actual seasonal load profiles for a resort island in the South China Sea" published in Energy (Khan et al., 2015) [1].

Event-related potentials (ERPs) time-locked to decision outcomes are reported. Participants engaged in a gambling task (see [1] for details] in which they decided between a risky and a safe option (presented as different coloured shapes) on each trial (416 in total). Each decision was accompanied with (fully randomised) feedback about the reward outcome (Win/Loss) and its magnitude (varying as a function of decision response; 5–9 points for Risky decisions and 1–4 points for Safe decisions). Here, we show data demonstrating: (a) The influence of Win feedback in the preceding outcome (Outcomet−1) on activity related to the current outcome (Outcomet); (b) Difference wave analysis for outcome expectancy- separating Expected Outcomes (consecutive loss trials subtracted from consecutive reward) from Unexpected Outcomes (subtracting Losst−1Wint trials from Wint−1Losst trials); (c) Difference waves separating Switch and Stay responses for Outcome Expectancy; (d) The effect of magnitude induced by decisions (Riskt vs. Safet) on Outcome Expectancy; and finally, (e) Expectations reflected by response switch direction (Risk to Safe responses vs. Safe to Riskt) on the FRN at Outcomet.

Development and validation of multiresidue analytical method in cotton and groundnut oil for 87 pesticides using low temperature and dispersive cleanup on gas chromatography and liquid chromatography-tandem mass spectrometry
Abstract
A method was developed and validated for the analysis of 87 pesticides in cotton and groundnut oil by GC with ECD and FPD detectors and LC-MS/MS. The extraction procedure based on QuEChERS followed by low-temperature freezing and dispersive cleanup steps was validated in two oil matrices for 87 pesticides of different classes. Linearity, expressed as coefficient of variation, was within the acceptable range. Of those tested, 77–83 and 77–89 % pesticides showed recoveries within the acceptable range of 70–120 % on LC-MS/MS in cottonseed oil and groundnut oil, respectively, at different spiking levels. In case of GC analysis, 63–65 and 53–82 % pesticides showed recoveries within the acceptable range of 70–120 % on GC in cottonseed oil and groundnut oil, respectively, at different spiking levels. The exceptions to these recoveries were the few organochlorines which consistently gave lower recoveries. Recovery factors can be employed while analysing these pesticides by this method as the results obtained were consistent in both oils. RSD was less than 20 % for most of the pesticides. The calculated limit of quantitation (LOQ) for most of the pesticides satisfies the maximum residue level (MRL) requirements as per European Union (EU) guidelines and Food Safety and Standards Authority of India (FSSAI).

William Shatner on Tinnitus: When He Developed It
Tinnitus is a condition that affects around 50 million people in the United States. It is a condition where a person frequently experience buzzing or ringing in the ears. Tinnitus has a tendency to get worse with age. It can also be a sign of an underlying health condition, such as high blood pressure. In some cases, tinnitus occurs as the result of an injury to the ear.
Anyone can develop tinnitus. William Shatner, an actor whose career has lasted over 50 years, is one of the many people who have struggled with tinnitus. He stated that his battle with tinnitus began when he was a star on the popular 1960s show "Star Trek".
He stated that he was filming an episode entitled "Arena." He said that he stood very close to a special effects explosion during the filming. William has stated that he believes the sound of the explosion is what caused his tinnitus.
William Shatner on Tinnitus: His Struggle with the Condition
William Shatner struggled with tinnitus for many years. He was bothered by the constant ringing in his ears. He stated that he was tormented by tinnitus, and there were days when he felt like he could not go on because the condition was so bothersome.
William Shatner on Tinnitus: Overcoming the Condition
William Shatner stated that his life changed for the better after he discovered the American Tinnitus Foundation. He describes the foundation as being a "Ray of Light." Williams said that he used a technique called tinnitus retraining therapy in order to overcome tinnitus. This technique involves the use of white noise.
The theory behind the use of white noise is that if you keep exposing yourself to white noise, then your brain will become accustomed to it. This training also helps your brain get adjusted to tinnitus so that you will forget that you even have it. William said that he noticed a 95 percent reduction in his symptoms after he used tinnitus retraining therapy.
Even though there are millions of people in America who have tinnitus, they often feel like they are left to suffer in silence. William Shatner has teamed up with the American Tinnitus Foundation to raise awareness about this condition and to let people know that they are not alone. The American Tinnitus Foundation is also working towards finding a cure for this condition.

Mice lack of LRG-47 display the attenuated outcome of infection with Schistosoma japonicum
Abstract
Interferon-inducible GTPase LRG-47 (also named immune-related GTPase M, Irgm1) is a member of the p47 GTPase family that has been shown to regulate host resistance to intracellular pathogens. Little knowledge has been known about the role of LRG-47 in host's responses to extracellular pathogens. To investigate possible roles of LRG-47 in the course of Schistosoma japonicum infection, LRG-47-deficient (LRG-47−/−) and wild-type (WT) mice were challenged with cercariae of S. japonicum, and the cellular and humoral responses in mice were analyzed. At the acute stage of S. japonicum infection, in contrast to WT mice, LRG-47−/− mice showed the significantly decreased egg burden, low schistosome-specific antibody response, and the decreased Th1 and increased Tc1 responses. Additionally, Schistosoma japonicum-specific egg antigen immunization also produced the similar humoral and cellular immune responses as S. japonicum infection. Taken together, these data suggested that the deficiency of LRG-47 might affect host's CD4+ T cell immune response via the weakening of IFN-γ downstream signaling; however, the specific function of LRG-47 on dealing with extracellular worm needs to be further studied.

Anthelmintic effects of Oroxylum indicum stem bark extract on juvenile and adult stages of Hymenolepis diminuta (Cestoda), an in vitro and in vivo study
Abstract
Worldwide, traditional usage of herbal medicines is a common practice to treat various parasitic infections. In India, bark decoction of Oroxylum indicum (L.) Kurz. (Bignoniaceae) is used as a traditional medicine to cure intestinal-helminthic infections. This study investigated the anthelmintic efficacy of methanolic bark extract of O. indicum on Hymenolepis diminuta (Cestoda), using both in vitro and in vivo methods. Utilizing a mini-questionnaire, first, we collected information about the pattern of anthelmintic use of this plant. Later, in vitro efficacy of extract was tested at 10, 20 and 30 mg/ml on both the artificially excysted juveniles and adult H. diminuta worms. Herein, scanning electron microscopy (SEM) was also utilized to determine the possible effects of extract on tegumental surfaces of juvenile and adult cestode. In vivo, extract was tested at 250, 500 and 1000 mg/kg in H. diminuta albino rat model, against juvenile and adult cestode. Praziquantel (PZQ) served as reference drug in anthelmintic assays. The acute toxicity of extract was determined as per the OECD guidelines. The field questionnaire data revealed that 78 % of people in the area use O. indicum stem bark against intestinal helminths, and of these, 75 % of people also believed it highly efficacious anthelmintic remedy. In vitro testing of extract revealed significant effects on juvenile worms, and 30 mg/ml of extract caused mortality of juveniles at the initial period (0.25 ± 0.00 h). Conversely, PZQ (1 mg/ml) showed paralysis and mortality of juvenile cestodes in 0.44 ± 0.04 and 1.11 ± 0.06 h, respectively. As determined by SEM, in vitro exposure to extract showed substantial effects on both juveniles and adult worms in the form of wrinkled scolex, distorted tegument and eroded microtriches. In vivo study revealed better efficacy of extract against juveniles than adult stages of parasite. Treatment of rats with 1000 mg/kg of extract caused 79.3 % reduction in EPG counts and 70.8 % of reduction in worm counts, which compared well to PZQ efficacy. In acute toxicity study, administration of a single oral dose of 2000 mg/kg of extract to mice did not cause any signs of toxicity or mortality within 2-week observation. Hence, the LD50 of extract was estimated to be greater than 2000 mg/kg. In conclusion, the bark extract of O. indicum possesses concentration-dependent effects against the larval and adult H. diminuta worms, and thus the extract holds promise in the control of intestinal helminthiases. This also supports the common belief of people about the reputation of this plant as anthelmintic in traditional medicine. There is a need to standardize the formulations of this plant for their possible use as standardized herbal anthelmintic agents in the future.

Morphological and molecular characterization of Myxobolus mucosus sp. n. (Myxosporea: Myxobolidae) with basifilamental sporulation in two cyprinid fishes, Rutilus rutilus (L.) and Leuciscus leuciscus (L.) in Russia
Abstract
A new Myxobolus species, Myxobolus mucosus sp. n., was found in the basifilamental location of roach, Rutilus rutilus, and common dace, Leuciscus leuciscus, during the investigation of fish myxosporean fauna in Finnish Bay of Baltic Sea and Lake Ladoga in 2014. Plasmodia from the two hosts share similar morphometric and morphological features which are whitish and slightly elongated oval and measured 300–500 μm in diameter. Mature spores of M. mucosus sp. n. are ellipsoidal in frontal view and lemon shaped in sutural view, measuring 13.6 ± 0.62 (12.8–14.2) μm in length, 10.7 ± 0.51 (10.3–11.4) μm in width, 7.8 ± 0.41 (7.2–8.3) μm in thickness and 13.0 ± 0.89 (11.5–14.2) μm in length, 10.8 ± 0.77 (10.2–11.5) μm in width, 7.7 ± 0.52 (7.3–8.0) μm in thickness from roach and common dace, respectively. Polar capsules were pyriform, equal in size, converging anteriorly, with 5.9 ± 0.22 (5.7–6.3) μm in length and 3.9 ± 0.3 (3.5–4.1) μm in width and 5.6 ± 0.38 (5.1–6.2) μm in length and 3.8 ± 0.24 (3.5–4.0) μm in width from roach and common dace, respectively. Spore valves are relatively thin, symmetrical, and smooth. The most remarkable morphological character of this new Myxobolus species is the distinct mucous envelope, surrounding the full myxospore valves, up to 25 and 32 μm, respectively, in roach and common dace. The partial 18S ribosomal DNA (rDNA) sequence variation of myxospores from two hosts is very tiny (below 0.8 %) but significantly differing from all available sequences of myxosporeans in GenBank. Therefore, it can be concluded that this myxosporean from these two closely related cyprinids are conspecific by combining morphological characteristics, tissue specificity, and molecular data. Phylogenetic analysis showed that this newly described species cluster with several gill-infecting Myxobolus species from cyprinid fish inhabiting in Eastern Europe.

Best practices for assessing ocean health in multiple contexts using tailorable frameworks
Marine policy is increasingly calling for maintaining or restoring healthy oceans while human activities continue to intensify. Thus, successful prioritization and management of competing objectives requires a comprehensive assessment of the current state of the ocean. Unfortunately, assessment frameworks to define and quantify current ocean state are often site-specific, limited to a few ocean components, and difficult to reproduce in different geographies or even through time, limiting spatial or temporal comparisons as well as the potential for shared learning. Ideally, frameworks should be tailorable to accommodate use in disparate locations and contexts, removing the need to develop frameworks de novo and allowing efforts to focus on the assessments themselves to advise action. Here, we present some of our experiences using the Ocean Health Index (OHI) framework, a tailorable and repeatable approach that measures health of coupled human-ocean ecosystems in different contexts by accommodating differences in local environmental characteristics, cultural priorities, and information availability and quality. Since its development in 2012, eleven assessments using the OHI framework have been completed at global, national, and regional scales, four of which have been led by independent academic or government groups. We have found the following to be best practices for conducting assessments: Incorporate key characteristics and priorities into the assessment framework design before gathering information; Strategically define spatial boundaries to balance information availability and decision-making scales; Maintain the key characteristics and priorities of the assessment framework regardless of information limitations; and Document and share the assessment process, methods, and tools. These best practices are relevant to most ecosystem assessment processes, but also provide tangible guidance for assessments using the OHI framework. These recommendations also promote transparency around which decisions were made and why, reproducibility through access to detailed methods and computational code, repeatability via the ability to modify methods and computational code, and ease of communication to wide audiences, all of which are critical for any robust assessment process.

Mortality (1968-2008) in a French cohort of uranium enrichment workers potentially exposed to rapidly soluble uranium compounds
Objectives
Until recently, enrichment of uranium for civil and military purposes in France was carried out by gaseous diffusion using rapidly soluble uranium compounds. We analysed the relationship between exposure to soluble uranium compounds and exposure to external -radiation and mortality in a cohort of 4688 French uranium enrichment workers who were employed between 1964 and 2006.
MethodsData on individual annual exposure to radiological and non-radiological hazards were collected for workers of the AREVA NC, CEA and Eurodif uranium enrichment plants from job-exposure matrixes and external dosimetry records, differentiating between natural, enriched and depleted uranium. Cause-specific mortality was compared with the French general population via standardised mortality ratios (SMR), and was analysed via Poisson regression using log-linear and linear excess relative risk models.
ResultsOver the period of follow-up, 131 161 person-years at risk were accrued and 21% of the subjects had died. A strong healthy worker effect was observed: all causes SMR=0.69, 95% CI 0.65 to 0.74. SMR for pleural cancer was significantly increased (2.3, 95% CI 1.06 to 4.4), but was only based on nine cases. Internal uranium and external -radiation exposures were not significantly associated with any cause of mortality.
ConclusionsThis is the first study of French uranium enrichment workers. Although limited in statistical power, further follow-up of this cohort, estimation of internal uranium doses and pooling with similar cohorts should elucidate potential risks associated with exposure to soluble uranium compounds.

A case-control study of mesothelioma in Minnesota iron ore (taconite) miners
Objectives
An excess of mesothelioma has been observed in iron ore miners in Northeastern Minnesota. Mining and processing of taconite iron ore generate exposures that include elongate mineral particles (EMPs) of amphibole and non-amphibole origin. We conducted a nested case–control study of mesothelioma in a cohort of 68 737 iron ore miners (haematite and taconite ore miners) to evaluate the association between mesothelioma, employment and EMP exposures from taconite mining.
MethodsMesothelioma cases (N=80) were identified through the Minnesota Cancer Surveillance System (MCSS) and death certificates. Four controls of similar age were selected for each case with 315 controls ultimately eligible for inclusion. Mesothelioma risk was evaluated by estimating rate ratios and 95% CIs with conditional logistic regression in relation to duration of taconite industry employment and cumulative EMP exposure [(EMP/cc)xyears], defined by the National Institute for Occupational Safety and Health (NIOSH) 7400 method. Models were adjusted for employment in haematite mining and potential exposure to commercial asbestos products used in the industry.
ResultsAll mesothelioma cases were male and 57 of the cases had work experience in the taconite industry. Mesothelioma was associated with the number of years employed in the taconite industry (RR=1.03, 95% CI 1.00 to 1.06) and cumulative EMP exposure (RR=1.10, 95% CI 0.97 to –1.24). No association was observed with employment in haematite mining.
ConclusionsThese results support an association between mesothelioma and employment duration and possibly EMP exposure in taconite mining and processing. The type of EMP was not determined. The potential role of commercial asbestos cannot be entirely ruled out.

The Lipidomic Analyses in Low and Highly Aggressive Ovarian Cancer Cell Lines
Abstract
Despite huge advances in the research of epithelial ovarian cancer (EOC), it remains the most lethal gynecological malignancy. Peritoneal tumor cell dissemination with cell survival and drug-resistance to taxane and platinum-based chemotherapy are two of the major challenges of EOC treatment. We have generated highly aggressive EOC cell lines (ID8-P1 lines or P1) from ID8-P0 (without in vivo passage, or P0) through in vivo passage in mice. We conducted lipidomic analyses in cells from ID8-P0 versus three ID8-P1 cell lines using ultra-high-performance liquid chromatography coupled to electrospray ionization tandem mass spectrometry. A total of 16 classes of lipids (149 individual lipids) were analyzed and compared between P0 and P1 cells. In addition to overall lipid profiles in EOC cells, we had several novel observations. Several classes and species of lipids have been identified to be differentially present in P0 versus P1 cells, which are potentially involved in the acquired aggressiveness of P1 cells. Triacylglycerols (TAG) were dramatically increased under detachment stress in EOC cells. Since survival of EOC cells under detachment is one of the major obstacles for EOC treatment, further studies identifying the molecular mechanisms controlling TAG increase may lead to new treatment modalities for EOC.

Preoperative versus postoperative ultrasound-guided rectus sheath block for improving pain, sleep quality and cytokine levels of patients with open midline incisions undergoing transabdominal gynaecological operation: study protocol for a randomised controlled trial
Background: Rectus sheath block (RSB) is used for postoperative pain relief in patients undergoing abdominal surgery with midline incision. Preoperative RSB has been shown to be effective, but it has not been compared with postoperative RSB. The aim of the present study is to evaluate postoperative pain, sleep quality and changes in the cytokine levels of patients undergoing gynaecological surgery with RSB performed preoperatively versus postoperatively.Methods/DesignThis study is a prospective, randomised, controlled (randomised, parallel group, concealed allocation), single-blinded trial. All patients undergoing transabdominal gynaecological surgery will be randomised 1:1 to the treatment intervention with general anaesthesia as an adjunct to preoperative or postoperative RSB. The objective of the trial is to evaluate postoperative pain, sleep quality and changes in the cytokine levels of patients undergoing gynaecological surgery with RSB performed preoperatively (n = 32) versus postoperatively (n = 32). All of the patients, irrespective of group allocation, will receive patient-controlled intravenous analgesia (PCIA) with oxycodone.The primary objective is to compare the interval between leaving the post-anaesthesia care unit and receiving the first PCIA bolus injection on the first postoperative night between patients who receive preoperative versus postoperative RSB. The secondary objectives will be to compare (1) cumulative oxycodone consumption at 24 hours after surgery; (2) postoperative sleep quality, as measured using a BIS-Vista monitor during the first night after surgery; and (3) cytokine levels (interleukin-1, interleukin-6, tumour necrosis factor-α and interferon-γ) during surgery and at 24 and 48 hours postoperatively.DiscussionClinical experience has suggested that RSB is a very effective postoperative analgesic technique, and we will answer the following questions with this trial. Do preoperative block and postoperative block have the same duration of analgesic effects? Can postoperative block extend the analgesic time? The results of this study could have actual clinical applications that could help to reduce postoperative pain and shorten hospital stays.Trial registrationCurrent Controlled Trials NCT02477098 15 June 2015.

Comparison of total ankle replacement and ankle arthrodesis in patients with haemophilia using gait analysis: two case reports
Background: Severe hemophilia is an inherited, lifelong bleeding disorder characterized by spontaneous bleeding, which results in painful joint deformities. Currently two surgical treatments are available to treat haemophilia-related ankle joint destruction: ankle arthrodesis and total ankle replacement. The aim of the present study was to compare these two surgical procedures in haemophiliac subjects.Case presentationKinematic and dynamic parameters were quantified using a three-dimensional gait-analysis system in two similar clinical cases. In Caucasian case 1, ankle arthrodesis was chosen because of a kinematic ankle flexion defect and lack of dynamic power regeneration. The defect in energy absorption was compensated for by the contralateral side. Total ankle replacement in Caucasian case 2 allowed sparing the ipsilateral knee (maximum 0.27 preoperatively vs. 0.71 W/kg postoperatively) and hip joints powers (maximum 0.43 preoperatively vs. 1.25 W/kg postoperatively) because of the small ankle dorsiflexion motion. Conclusions: Total ankle replacement is recommended for haemophiliac patients who present with a preserved ankle range of motion.

Detection of mcr-1 encoding plasmid-mediated colistin-resistant Escherichia coli isolates from human bloodstream infection and imported chicken meat, Denmark 2015
The plasmid-mediated colistin resistance gene, mcr-1, was detected in an Escherichia coli isolate from a Danish patient with bloodstream infection and in five E. coli isolates from imported chicken meat. One isolate from chicken meat belonged to the epidemic spreading sequence type ST131. In addition to IncN2, an incX4 replicon was found to be linked to mcr-1. This report follows a recent detection of mcr-1 in E. coli from animals, food and humans in China.

Colonisation with toxigenic Corynebacterium diphtheriae in a Scottish burns patient, June 2015
On 12 June 2015, Corynebacterium diphtheriae was identified in a skin swab from a burns patient in Scotland. The isolate was confirmed to be genotypically and phenotypically toxigenic. Multilocus sequence typing of three patient isolates yielded sequence type ST 125. The patient was clinically well. We summarise findings of this case, and results of close contact identification and screening: 12 family and close contacts and 32 hospital staff have been found negative for C. diphtheriae.

Urine is a novel source of autologous mesenchymal stem cells for patients with epidermolysis bullosa
Background: Regenerative medicine is strictly dependent on stem cells as a source for a high diversity of somatic cells. However, the isolation of such from individuals suffering from severe genetic skin blistering diseases like epidermolysis bullosa (EB) is often associated with further organ damage. Methods: Stem cells were isolated from 112 urine samples from 21 different healthy donors, as well as from 33 urine samples from 25 donors with EB. The cultivation of these cells was optimized by testing different media formulations and pre-coating of culture vessels with collagen. The identity of cells was confirmed by testing marker expression, differentiation potential and immune-modulatory properties. Results: We provide here an optimized protocol for the reproducible isolation of mesenchymal stem cells from urine, even from small volumes as obtained from patients with EB. Furthermore, we offer a basic characterization of those urine-derived stem cells (USCs) from healthy donors, as well as from patients with EB, and demonstrate their potential to differentiate into chondrocytes, osteoblasts and adipocytes, as well as their immune-modulatory properties. Conclusions: Thus, USCs provide a novel and non-invasive source of stem cells, which might be applied for gene-therapeutic approaches to improve medical conditions of patients with EB.

The purpose of this study was to quantify strain and elongation of the long head of the biceps femoris (BFlh) and the semitendinosus (ST) tendon/aponeurosis. Forty participants performed passive knee extension trials from 90° of knee flexion to full extension (0°) followed by ramp isometric contractions of the knee flexors at 0, 45 and 90° of knee flexion. Two ultrasound probes were used to visualize the displacement of BFlh and ST tendon/aponeurosis. Three-way analysis of variance designs indicated that: (a) Tendon/aponeurosis (passive) elongation and strain were higher for the BFlh than the ST as the knee was passively extended (p<0.05) (b) Contraction at each angular position was accompanied by a smaller BFlh tendon/aponeurosis (active) strain and elongation than the ST at higher levels of effort (p <0.05) and c) Combined (passive and active) strain was significantly higher for the BFlh than ST during ramp contraction at 0° but the opposite was observed for the 45° and 90° flexion angle tests (p < 0.05). Passive elongation of tendon/aponeurosis has an important effect on the tendon/aponeurosis behavior of the hamstrings and may contribute to a different loading of muscle fibers and tendinous tissue between BFlh and ST.

Previous studies have shown an association of visual demands during near work and increased activity of the trapezius muscle. Those studies were conducted under stationary postural conditions with fixed gaze and artificial visual load. The present study investigated the relationship between ciliary muscle contraction force and trapezius muscle activity across individuals during performance of a natural dynamic motor task under free gaze conditions. Participants (N=11) tracked a moving visual target with a digital pen on a computer screen. Tracking performance, eye refraction and trapezius muscle activity were continuously measured. Ciliary muscle contraction force was computed from eye accommodative response. There was a significant Pearson correlation between ciliary muscle contraction force and trapezius muscle activity on the tracking side (0.78, p<0.01) and passive side (0.64, p<0.05). The study supports the hypothesis that high visual demands, leading to an increased ciliary muscle contraction during continuous eye-hand coordination, may increase trapezius muscle tension and thus contribute to the development of musculoskeletal complaints in the neck-shoulder area. Further experimental studies are required to clarify whether the relationship is valid within each individual or may represent a general personal trait, when individuals with higher eye accommodative response tend to have higher trapezius muscle activity.
