Resolution Optimization Based on Fluorophore Photophysics in Single-Molecule Localization Microscopy

Since their advent nearly a decade ago, superresolution microscopy techniques based on single-molecule fluorescence (1–3) have substantially expanded our knowledge of subcellular organization and dynamics. By pushing spatial resolutions well below the diffraction limit, single-molecule superresolution microscopy allows researchers to peer into individual cells and examine the intricate architecture and biochemical events on small size scales previously inaccessible through conventional light microscopy.
