Development of analytical methodologies to assess recalcitrant pesticide bioremediation in biobeds at laboratory scale

To assess recalcitrant pesticide bioremediation it is necessary to gradually increase the complexity of the biological system used in order to design an effective biobed assembly. Each step towards this effective biobed design needs a suitable, validated analytical methodology that allows a correct evaluation of the dissipation and bioconvertion. Low recovery yielding methods could give a false idea of a successful biodegradation process. To address this situation, different methods were developed and validated for the simultaneous determination of endosulfan, its main three metabolites, and chlorpyrifos in increasingly complex matrices where the bioconvertor basidiomycete Abortiporus biennis could grow. The matrices were culture media, bran, and finally a laboratory biomix composed of bran, peat and soil. The methodology for the analysis of the first evaluated matrix has already been reported. The methodologies developed for the other two systems are presented in this work. The targeted analytes were extracted from fungi growing over bran in semisolid media YNB (Yeast Nitrogen Based) with acetonitrile using shaker assisted extraction, The salting-out step was performed with MgSO4 and NaCl, and the extracts analyzed by GC-ECD. The best methodology was fully validated for all the evaluated analytes at 1 and 25mgkg−1 yielding recoveries between 72% and 109% and RSDs <11% in all cases. The application of this methodology proved that A. biennis is able to dissipate 94% of endosulfan and 87% of chlorpyrifos after 90 days. Having assessed that A. biennis growing over bran can metabolize the studied pesticides, the next step faced was the development and validation of an analytical procedure to evaluate the analytes in a laboratory scale biobed composed of 50% of bran, 25% of peat and 25% of soil together with fungal micelium. From the different procedures assayed, only ultrasound assisted extraction with ethyl acetate allowed recoveries between 80% and 110% with RSDs <18%. Linearity, recovery, precision, matrix effect and LODs/LOQs of each method were studied for all the analytes: endosulfan isomers (α & β) and its metabolites (endosulfan sulfate, ether and diol) as well as for chlorpyrifos. In the first laboratory evaluation of these biobeds endosulfan was bioconverted up to 87% and chlorpyrifos more than 79% after 27 days.

Enhancement of periodate-hydrogen peroxide chemiluminescence by nitrogen doped carbon dots and its application for the determination of pyrogallol and gallic acid

A new sensitized chemiluminescence (CL) was developed to broaden the analytical application of KIO4–H2O2 system. The nitrogen doped carbon dots (N-CDs) dramatically boosted the CL intensity of KIO4–H2O2 system which was further enriched by basic medium. In light of EPR analysis, free radical scavenging studies and CL spectra the detail mechanism for the enhancement was conferred in the presence of N-CDs and NaOH. The results suggested that CL of KIO4–H2O2 system in the presence and absence of N-CDs and NaOH proceeds via radical pathway. The enhanced CL was used for the determination of pyrogallol and gallic acid in range of 1.0×10−4–1.0×10−7M with 4.6×10−8 and 6.1×10−8M limit of detection respectively. The relative standard deviation (RSD) at a concentration of 10−5 for gallic acid and pyrogallol was 1.4% and 2.3% respectively (n=11). The attained results unveil that the present method is sensitive, faster, simpler and less costly compared to other methods and could be applied to determine polyphenols in real samples.

Sequential morphological change of Chiari malformation type II following surgical repair of myelomeningocele

Abstract

Purpose

To document long-term morphological changes of Chiari type II malformation (CM-II) following closure of spina bifida manifesta (SBM).

Methods

We retrospectively evaluated postnatal magnetic resonance images of the CM-II and posterior fossa (PF) in 28 consecutive cases. We measured changes in vertebral level and length of the cerebellar peg (CP), cerebrospinal fluid (CSF) spaces anterior and posterior to the cerebrospinal junction, PF area, and the anteroposterior diameters of the foramen magnum (FM) and C1 vertebra. We examined the morphological differences between the cases with and without ventriculoperitoneal (VP) shunting and derived predicted means by nonlinear mixed-effect modeling.

Results

At birth, there were significant differences in CP length, PF area, and FM and C1 diameters between those who underwent VP shunting and those who did not. In cases with a CP below C1, VP shunting was required in every case but one. In those with visible CSF space at birth, VP shunts were not required. In 17 of 18 cases with a CP below C1, the vertebral level ascended by mean two vertebral levels (range 0–5 levels) within 4–6 months of delivery. In the remaining case, slowly progressive hydrocephalus and delayed CP descent required VP shunting at 8 months. Predicted mean CP length and FM and C1 diameters were greater in those who underwent VP shunting, but there was no difference in predicted mean PF area.

Conclusion

The morphology of CM-II and the presence of hydrocephalus influence each other in children who have undergone postnatal SBM repair.

Toxins, Vol. 8, Pages 64: Isolation of Anti-Ricin Protective Antibodies Exhibiting High Affinity from Immunized Non-Human Primates

Ricin, derived from the castor bean plant Ricinus communis, is one of the most potent and lethal toxins known, against which there is no available antidote. To date, the use of neutralizing antibodies is the most promising post-exposure treatment for ricin intoxication. The aim of this study was to isolate high affinity anti-ricin antibodies that possess potent toxin-neutralization capabilities. Two non-human primates were immunized with either a ricin-holotoxin- or subunit-based vaccine, to ensure the elicitation of diverse high affinity antibodies. By using a comprehensive set of primers, immune scFv phage-displayed libraries were constructed and panned. A panel of 10 antibodies (five directed against the A subunit of ricin and five against the B subunit) was isolated and reformatted into a full-length chimeric IgG. All of these antibodies were found to neutralize ricin in vitro, and several conferred full protection to ricin-intoxicated mice when given six hours after exposure. Six antibodies were found to possess exceptionally high affinity toward the toxin, with KD values below pM (koff < 1 × 10−7 s−1) that were well correlated with their ability to neutralize ricin. These antibodies, alone or in combination, could be used for the development of a highly-effective therapeutic preparation for post-exposure treatment of ricin intoxication.

Chitosan is a biopolymer with a wide range of applications. The use of chitosan in clinical medicine to control infections by fungal pathogens such as Candida spp. is one of its most promising applications in view of the reduced number of antifungals available. Chitosan increases intracellular oxidative stress, then permeabilizes the plasma membrane of sensitive filamentous fungus Neurospora crassa and yeast. Transcriptomics reveals plasma membrane homeostasis and oxidative metabolism genes as key players in the response of fungi to chitosan. A lipase and a monosaccharide transporter, both inner plasma membrane proteins, and a glutathione transferase are main chitosan targets in N. crassa. Biocontrol fungi such as Pochonia chlamydosporia have a low content of polyunsaturated free fatty acids in their plasma membranes and are resistant to chitosan. Genome sequencing of P. chlamydosporia reveals a wide gene machinery to degrade and assimilate chitosan. Chitosan increases P. chlamydosporia sporulation and enhances parasitism of plant parasitic nematodes by the fungus. Omics studies allow understanding the mode of action of chitosan and help its development as an antifungal and gene modulator.

Toxins, Vol. 8, Pages 63: Characterization and Recombinant Expression of Terebrid Venom Peptide from Terebra guttata

Venom peptides found in terebrid snails expand the toolbox of active compounds that can be applied to investigate cellular physiology and can be further developed as future therapeutics. However, unlike other predatory organisms, such as snakes, terebrids produce very small quantities of venom, making it difficult to obtain sufficient amounts for biochemical characterization. Here, we describe the first recombinant expression and characterization of terebrid peptide, teretoxin Tgu6.1, from Terebra guttata. Tgu6.1 is a novel forty-four amino acid teretoxin peptide with a VI/VII cysteine framework (C–C–CC–C–C) similar to O, M and I conotoxin superfamilies. A ligation-independent cloning strategy with an ompT protease deficient strain of E. coli was employed to recombinantly produce Tgu6.1. Thioredoxin was introduced in the plasmid to combat disulfide folding and solubility issues. Specifically Histidine-6 tag and Ni-NTA affinity chromatography were applied as a purification method, and enterokinase was used as a specific cleavage protease to effectively produce high yields of folded Tgu6.1 without extra residues to the primary sequence. The recombinantly-expressed Tgu6.1 peptide was bioactive, displaying a paralytic effect when injected into a Nereis virens polychaete bioassay. The recombinant strategy described to express Tgu6.1 can be applied to produce high yields of other disulfide-rich peptides.

The aim of this review is to understand how physical activity and nutrition are involved in the improvement of sarcopenia in patients affected by liver cirrhosis. The pathogenesis of sarcopenia in cirrhosis involves three major factors: inadequate dietary intake, metabolic disturbances, and malabsorption. Although in the early stages muscles appear to be spared, sarcopenia progressively leads to mobility limitations and its consequences, such as propensity to falls and drastically reducing life quality. Several studies confirm the important role played by physical activity and balanced nutrition in this chronic condition. Exercise and nutritional intervention should be recommended in these patients in order to improve quality of life.

Energies, Vol. 9, Pages 155: Adaptive Procurement Guidelines for Automatic Selection of Renewable Forest Energy Sources within a Sustainable Energy Production System

An automatic forest-stand selection method was developed that integrates the procurement of profitable energy sources within a sustainable energy production system. We tested the method using a forest harvester simulator. We found that site-specific estimates of forest characteristics are important when predicting the potential of an energy-wood stand as a renewable energy source. Further, tree parameters can be combined with automatic stem measurements from a multiple-tree harvester to predict the energy-wood biomass in the stands. The selection process uses data from profitability studies. The selection process also uses environmental criteria to ensure that sufficient soil organic matter is left behind and to protect the soil against erosion. The integrated system of the harvester automatically adapts the system's models and stand-selection rules to account for various site-specific stand parameters. Predicting the profitable and environmentally acceptable yield of stand biomass has great potential in sustainable forest resource management, but managers must decide whether the operational procurement guidelines provided by the stand-selection method is acceptable under their local real-world wood procurement conditions.

Sustainability, Vol. 8, Pages 237: Characteristics of Land Use/Cover and Macroscopic Ecological Changes in the Headwaters of the Yangtze River and of the Yellow River over the Past 30 Years

Based on land use and land cover (LULC) datasets in the late 1970s, the early 1990s, 2004 and 2012, we analyzed characteristics of LULC change in the headwaters of the Yangtze River and Yellow River over the past 30 years contrastively, using the transition matrix and LULC change index. The results showed that, in 2012, the LULC in the headwaters of the Yellow River were different compared to those of the headwaters of the Yangtze River, with more grassland and wet- and marshland. In the past 30 years, the grassland and wet- and marshland increasing at the expense of sand, gobi, and bare land and desert were the main LULC change types in the headwaters of the Yangtze River, with the macro-ecological situation experiencing a process of degeneration, slight melioration, and continuous melioration, in that order. In the headwaters of the Yellow River, severe reduction of grassland coverage, shrinkage of wet- and marshland and the consequential expansion of sand, gobi and bare land were noticed. The macro-ecological situation experienced a process of degeneration, obvious degeneration, and slight melioration, in that order, and the overall change in magnitude was more dramatic than that in the headwaters of the Yangtze River. These different LULC change courses were jointly driven by climate change, grassland-grazing pressure, and the implementation of ecological construction projects.

Mother–offspring recognition in the domestic cat: Kittens recognize their own mother's call

ABSTRACT

Acoustic communication can play an important part in mother-young recognition in many mammals. This, however, has still only been investigated in a small range mainly of herd- or colony-living species. Here we report on the behavioral response of kittens of the domestic cat, a typically solitary carnivore, to playbacks of "greeting chirps" and "meows" from their own versus alien mothers. We found significantly stronger responses to the chirps from kittens' own mother than to her meows or to the chirps or meows of alien mothers. Acoustic analysis revealed greater variation between vocalizations from different mothers than for vocalizations from the same mother. We conclude that chirps emitted by mother cats at the nest represent a specific form of vocal communication with their young, and that kittens learn and respond positively to these and distinguish them from chirps of other mothers and from other cat vocalizations while still in the nest. © 2016 Wiley Periodicals, Inc. The textile industry achieves economic benefits through outsourcing to low cost markets. Today, reshoring is an emerging trend due to rising cost and unemployment concerns. This problem is primarily due to an industry-wide focus on economic benefits only. Cost saving is a basic reason for international outsourcing while domestic outsourcing provides capacity flexibility. Moreover, outsourcing risk has a major impact on strategic location of the production destinations. Therefore, the merging of capacity flexibility and outsourcing risk comprises a sustainable outsourcing strategy. This paper suggests a sustainable outsourcing strategy in which a textile manufacturer outsources to international markets for cost savings and outsources to the domestic market for capacity flexibility. The manufacturer reserves some capacity with domestic suppliers, and pays a unit penalty cost if this capacity flexibility is not utilized. The manufacturer seeks minimum risk in international markets. Operational cost, penalty cost, and outsourcing risk are considered to be objective functions. Decisions include the assignment of contracts to suitable facilities, the quantity of each contract, and allocation of reserved capacity flexibility among domestic suppliers. Multi-objective problem of this research was solved using three variants of goal programming. Several insights are proposed for outsourcing decision making in the current global environment.

Cancers, Vol. 8, Pages 31: Differentially Expressed MicroRNAs in Meningiomas Grades I and II Suggest Shared Biomarkers with Malignant Tumors

Meningiomas represent the most common primary tumors of the central nervous system, but few microRNA (miRNA) profiling studies have been reported so far. Deep sequencing of small RNA libraries generated from two human meningioma biopsies WHO grades I (benign) and II (atypical) were compared to excess dura controls. Nineteen differentially expressed miRNAs were validated by RT-qPCR using tumor RNA from 15 patients and 5 meninges controls. Tumor suppressor miR-218 and miR-34a were upregulated relative to normal controls, however, miR-143, miR-193b, miR-451 and oncogenic miR-21 were all downregulated. From 10 selected putative mRNA targets tested by RT-qPCR only four were differentially expressed relative to normal controls. PTEN and E-cadherin (CDH1) were upregulated, but RUNX1T1 was downregulated. Proliferation biomarker p63 was upregulated with nuclear localization, but not detected in most normal arachnoid tissues. Immunoreactivity of E-cadherin was detected in the outermost layer of normal arachnoids, but was expressed throughout the tumors. Nuclear Cyclin D1 expression was positive in all studied meningiomas, while its expression in arachnoid was limited to a few trabecular cells. Meningiomas of grades I and II appear to share biomarkers with malignant tumors, but with some additional tumor suppressor biomarkers expression. Validation in more patients is of importance.

Water, Vol. 8, Pages 78: Overland Transport of Rotavirus and the Effect of Soil Type and Vegetation

Soil and vegetation are two critical factors for controlling the overland transport kinetics of pathogens in a natural environment. With livestock operations moving more towards concentrated animal operations, the need to dispose of a very large amount of manure in a localized area is becoming increasingly important. Animal manure contains a substantial amount of microbial pathogens, including rotavirus, which may pose a threat of contamination of water resources. This study examined the kinetics of rotavirus in overland transport, with an overall objective of optimizing the design of best management practices, especially vegetative filter strips. The overland transport of rotavirus was studied using three soil types (Catlin silt-loam, Darwin silty-clay, Alvin fine sandy-loam), spanning the entire spectrum of typical Illinois soil textures. A 20-min rainfall event was produced using a small-scale (1.07 m × 0.66 m) laboratory rainfall simulator over a soil box measuring 0.610 m × 0.305 m. Each soil type was tested for rotavirus transport kinetics with bare surface conditions, as well as with Smooth Brome and Fescue vegetative covers. Surface runoff, near-surface runoff, soil cores, and vegetation were each analyzed for infective rotavirus particles using cell-culture infectivity assays. Results show that vegetation reduces the recovery of infective rotavirus particles in surface runoff by an average of 73%, in addition to delaying the time to peak recovery. The vegetation, in general, appeared to decrease the recovery of infective rotavirus particles in surface runoff by impeding surface flow and increasing the potential for infiltration into the soil profile.

Plant positive strand RNA viruses are intracellular infectious agents that take advantage of cellular lipids and membranes to support replication and protect viral RNA from degradation by host antiviral responses. In this review, we discuss how Tomato bushy stunt virus (TBSV) co-opts lipid transfer proteins and modulates lipid metabolism and transport to facilitate the assembly of the membrane-bound viral replicase complexes within intricate replication compartments. Identification and characterization of the proviral roles of specific lipids and proteins involved in lipid metabolism based on results from yeast (Saccharomyces cerevisiae) model host and cell-free approaches are discussed. The review also highlights the advantage of using liposomes with chemically defined composition to identify specific lipids required for TBSV replication. Remarkably, all the known steps in TBSV replication are dependent on cellular lipids and co-opted membranes.

The pine wood nematode, Bursaphelenchus xylophilus, causes huge economic losses in pine forests, has a complex life cycle, and shows the remarkable ability to survive under unfavorable and changing environmental conditions. This ability may be related to autophagy, which is still poorly understood in B. xylophilus and no autophagy-related genes have been previously characterized. In this study, transmission electron microscopy was used to confirm that autophagy exists in B. xylophilus. The full-length cDNAs of BxATG1 and BxATG8 were first cloned from B. xylophilus, and BxATG1 and BxATG8 were characterized using bioinformatics methods. The expression pattern of the autophagy marker BxATG8 was investigated using in situ hybridization (ISH). BxATG8 was expressed in esophageal gland and hypodermal seam cells. We tested the effects of RNA interference (RNAi) on BxATG1 and BxATG8. The results revealed that BxATG1 and BxATG8 were likely associated with propagation of nematodes on fungal mats. This study confirmed the molecular characterization and functions of BxATG1 and BxATG8 in B. xylophilus and provided fundamental information between autophagy and B. xylophilus.

Energies, Vol. 9, Pages 158: Rotor Design for an Efficient Single-Phase Induction Motor for Refrigerator Compressors

This article describes a rotor making technology for the production of high-efficiency single-phase induction motors (SPIMs) to be used in refrigerator compressors. Rotors can have different aluminum fill factors according to the fabrication method. In order to examine the association between the fill factor and the efficiency of the rotor, we analyzed the distribution of magnetic flux density using the finite element method (FEM). Next, we made prototype rotors by conventional casting methods and by the proposed casting method and compared their fill factors. In addition, SPIMs were made using the rotors, and their efficiencies were measured using a dynamometer. Sustainability, Vol. 8, Pages 228: An Evolutionary Approach to Adaptive Capacity Assessment: A Case Study of Soufriere, Saint Lucia

This paper assesses the capacity of Soufriere, Saint Lucia to adapt to climate change. A community-based vulnerability assessment was conducted that employed semi-structured interviews with community members. The results were analysed using the Local Adaptive Capacity (LAC) framework, which characterises adaptive capacity based on five elements: asset base; institutions and entitlements; knowledge and information; innovation; and flexible forward-looking decision-making and governance. Beyond providing an in-depth analysis of Soufriere's capacity to adapt to climate change, the paper argues that the elements of the LAC framework largely correspond with an evolutionary perspective on adaptive capacity. However, other evolutionary themes—such as structure, history, path-dependency, scale, agency, conservation of diversity, and the perils of specialisation—should also be taken into account.

Recycling, Vol. 1, Pages 136-146: Do Eco-Fees Encourage Design for the Environment? The Relationship between Environmental Handling Fees and Recycling Rates for Printed Paper and Packaging

This study undertook a critical examination of Ontario's extended producer responsibility scheme for the residential "Blue Box" recycling program, specifically examining the relationship between packaging fee rates and material-specific recycling rates. Using data collected for each of the 23 materials found in the residential recycling program over the past decade, a regression model was developed to gauge what relationship (if any) packaging recycling rates have with fee rates, costs of material management and revenue from the sale of recyclable material. The modeling in this study indicates that packaging fee rates have no effect on packaging recycling rates. Recycling rates were positively correlated with material revenue and negatively correlated with material management costs. There is no evidence that suggests that Ontario's fee model used to allocate environmental handling fees to individual materials encourages waste diversion or design for the environment. The disconnect in the results and the intended function of packaging fee rates calls into question the appropriateness of Ontario's fee rate methodology.

Water, Vol. 8, Pages 79: Strong Gradients in Weak Magnetic Fields Induce DOLLOP Formation in Tap Water

In 2012 Coey proposed a theory on the mechanism of magnetic water treatment based on the gradient of the applied field rather than its absolute strength. We tested this theory by measuring the effect of very weak field magnets (≤ 10 g) containing strong magnetic inhomogeneities (ΔB = 2 kg·m−1) on tap water samples by the use of electric impedance spectroscopy (EIS) and laser scattering. Our results show an increased formation of nm-sized prenucleation clusters (dynamically ordered liquid like oxyanion polymers or "DOLLOPs") due to the exposure to the magnetic field and thus are consistent with Coey's theory which is therefore also applicable to very weak magnetic fields as long as they contain strong gradients.

Materials, Vol. 9, Pages 149: Synthesis, Characterization and Cytotoxicity of Novel Multifunctional Fe3O4@SiO2@GdVO4:Dy3+ Core-Shell Nanocomposite as a Drug Carrier

In this study, multifunctional Fe3O4@SiO2@GdVO4:Dy3+ nanocomposites were successfully synthesized via a two-step method. Their structure, luminescence and magnetic properties were characterized by X-ray diffraction (XRD), scanning electronic microscope (SEM), transmission electron microscopy (TEM), photoluminescence (PL) spectra and vibrating sample magnetometer (VSM). The results indicated that the as-prepared multifunctional composites displayed a well-defined core-shell structure. The composites show spherical morphology with a size distribution of around 360 nm. Additionally, the composites exhibit high saturation magnetization (20.40 emu/g) and excellent luminescence properties. The inner Fe3O4 cores and the outer GdVO4:Dy3+ layers endow the composites with good responsive magnetic properties and strong fluorescent properties, which endow the nanoparticles with great potential applications in drug delivery, magnetic resonance imaging, and marking and separating of cells in vitro.
