Our front cover features immunofluorescent staining of thymic epithelial cells in an Ebi2−/− thymus, showing organization into cortical (pan-Cytokeratin, green) and medullary (Keratin5, red) regions. Post-positive selection thymocytes leave the cortex and enter the medulla where they migrate rapidly while scanning multiple antigen presenting cells displaying diverse self-antigens that induce central tolerance. This image is taken from Ki et al. (pp. 1906–1917), in which the authors found that EBI2 deficiency reduced the velocity of post-positive selection thymocytes in the medulla, resulting in impaired central tolerance induction.
